Here on macOS Sequoia, I had to run one more command to make it run:
chmod +x /YOUR-DIRECTORY-HERE/clonk.app/Contents/MacOS/clonk
(I believe this flags it as runnable for the OS)

Where did you download it?
The executable flag should definitely be set automatically.
It could be an issue on our side that we have to it.
Edit: I just checked the download from the table and it is indeed missing.
Edit 2: I just fixed it. Versions [361] to [363] were affected.
Also, installing nghttp2 is not necessary anymore since [363].
